- The subject of the public contract is the supply of sequencing chemistry for the Center of Excellence Creatic LF MU, specified in detail in the technical conditions of the contracting authority and carried out under the commercial conditions specified in Annex A of the procurement documentation. Masaryk University within its of instrumentation has instrumentation suitable for performing third-generation sequencing or long-read sequencing, namely the Revio System sequencer from Pacific Biosciences (PacBio), 1305 O'Brien Drive, Menlo Park, CA 94025, USA. For properly executed for long-read sequencing on this instrument, appropriate sequencing chemistry fully compatible with the existing instrument is necessary. For planned sequencing of samples using the Revio system sequencer, it is necessary to use reagents and appropriate sets/kits that are fully compatible with this system and ensure the correct functionality of the entire workflow. To ensure the correct functionality and reliability of the entire sequencing process, it is necessary to purchase reagents and kits that form a comprehensive and compatible system covering all steps of the workflow from isolation of nucleic acids, through the preparation of sequencing libraries, to the actual sequencing on the given device. The individual parts of this process are technologically connected and optimized so that they function as a whole. Change of supplier or manufacturer in some part (isolation, library preparation, sequencing chemistry) could lead to incompatibilities between reagents, reduced data quality, or failure of the sequencing run. For this reason, it is desirable (but not necessary) that reagents and kits used within a given workflow come from a single manufacturer or supplier that guarantees their mutual compatibility, validation and support for a specific type of Revio sequencing system.
